Guide Rock is a village on the Republican River in southern Webster County, named for a rocky bluff about 2.5 miles southeast of town that Pawnee people called Pa-Hur — "the hill that points the way" — and used for generations as a navigational landmark. The site held deep significance for the Pawnee long before American settlement: a large Kitkehahki-band Pawnee village stood near present-day Guide Rock from roughly the 1770s through the 1820s, and in 1806 it was visited first by a Spanish expedition and then, within weeks, by Lieutenant Zebulon Pike's American expedition, during which Pike is credited with persuading the Pawnee to lower the Spanish flag and raise the American one. The roughly 300-acre Pike-Pawnee Village Site was designated a National Historic Landmark in 1964 and added to the National Register of Historic Places in 1966; archaeological work there in the twentieth century helped establish Great Plains archaeology as a field of study.

The American town of Guide Rock traces to an 1870 settlement, with the town site itself surveyed and platted in 1873. The Burlington & Missouri River Railroad reached the area between 1878 and 1882, prompting the town to consolidate near the new tracks, and Guide Rock formally incorporated as a village in 1883, the same year its newspaper, the Guide Rock Signal, was founded. Guide Rock is the birthplace of screenwriter Lew Hunter (1935–2023), who spent decades as Chairman Emeritus and Professor of Screenwriting at UCLA, wrote for network television series including Bewitched and The Rockford Files, and mentored a generation of screenwriters — Steven Spielberg reportedly called him "the best screenwriting teacher going."

Guide Rock, incorporated in 1883, takes its name from a Pawnee landmark once used to navigate the Republican River valley; the nearby Pike-Pawnee Village Site, visited by Zebulon Pike's expedition in 1806, is a National Historic Landmark. The village is the birthplace of longtime UCLA screenwriting professor Lew Hunter.
