Wayne, founded in 1881 with the arrival of the Chicago, St. Paul, Minneapolis and Omaha Railway and named for Revolutionary War general "Mad Anthony" Wayne (the same namesake as the county), has grown into the county’s commercial and educational hub as home to Wayne State College. Its historic downtown commercial district is listed on the National Register of Historic Places.

As the county seat and by far the largest municipality in Wayne County — and one of the few in the county still gaining population — Wayne anchors the local economy through Wayne State College and its historic Main Street business district.
